目的:证明抗宫颈癌单克隆抗体(m Ab)AU14 - 1 识别的宫颈癌抗原决定簇是肿瘤相关基因表达的产物。方法:将宫颈癌(U14) 细胞DNA 转染原代培养正常淋巴细胞;分别用细胞和夹心酶联免疫吸附试验( ELISA) 检测DNA转染细胞膜表面抗原。结果:转染细胞如同U14 细胞一样对AU14 - 1 染色呈阳性反应;western 印迹法分析表明,转染细胞膜表面可溶性抗原呈现4 条与U14 细胞一致的分子量为20 000 、46 000 、61 000 和65 000 的蛋白多肽区带。结论:宫颈癌细胞膜表面抗原是肿瘤相关基因编码的低分子量糖蛋白。
PURPOSE: To demonstrate that cervical cancer epitopes recognized by anti-cervical cancer monoclonal antibody (mAb) AU14-1 are products of tumor-associated gene expression. Methods: DNA of cervical cancer (U14) was transfected into primary cultured normal lymphocytes. Cell surface and sandwich enzyme - linked immunosorbent assay (ELISA) were used to detect the DNA surface antigen. Results: The transfected cells were positive for AU14 - 1 staining as U14 cells. Western blot analysis showed that the soluble antigen on the surface of transfected cells showed four identical molecular weights with U14 cells of 20 000, 46 000, 61 000 and 65 000 protein polypeptide band. Conclusion: The cervical cancer cell surface antigen is a low molecular weight glycoprotein encoded by tumor-associated genes.
