目的分析吉林省2011年手足口病(hand,foot and mouth disease,HFMD)死亡病例标本中分离获得的肠道病毒71型(enterovirus 71,EV71)毒株全基因组基因特征。方法采用分段RT-PCR法对2011年吉林省HFMD死亡病例EV71分离株JL2011-56全基因组进行扩增和测序。利用Clustal X2和MEGA5软件构建分离株JL2011-56与Gen Bank中27株参考株的全基因组序列和VP1区全基因组序列的进化树,分析Jl2011-56株的流行病学特征;同时与Gen Bank中12株C4a基因亚型参考株进行全基因组核苷酸和氨基酸序列的同源性分析。结果 JL2011-56分离株属于C4a亚型,核苷酸序列和氨基酸序列与山东分离株SDLY107(JX244186.1)同源性最高,分别为99.2%和99.5%,与EV71原型株Br Cr的同源性分别为79.9%和95.7%。结论 JL2011-56分离株与山东株亲缘关系较近,可能存在共同的与中枢神经毒力相关的氨基酸突变位点。
Objective To analyze the genome-wide genomic characteristics of enterovirus 71 (EV71) isolated from the deaths of hand, foot and mouth disease (HFMD) in Jilin Province in 2011. Methods The segmented RT-PCR method was used to amplify and sequence the entire genome of J7120-56, the EV71 isolate of HFMD death in Jilin province in 2011. The Clustal X2 and MEGA5 software was used to construct the phylogenetic tree of the isolates JL2011-56 and 27 reference strains in Gen Bank and the VP1 genome-wide sequence to analyze the epidemiological characteristics of strain Jl2011-56. At the same time, Genome-wide nucleotide and amino acid sequence homology analysis of 12 C4a subtype reference strains. Results The JL2011-56 isolates belonged to C4a subtype. The nucleotide and amino acid sequences of JL2011-56 were 99.2% and 99.5% homologous to Shandong SDLY107 (JX244186.1), respectively. The homology Sexuality was 79.9% and 95.7% respectively. Conclusion The JL2011-56 isolate has a close genetic relationship with Shandong strain. There may be a common site of amino acid mutation associated with CNV virulence.
