目的研究原发性肝癌(HCC)患者肝组织中 HCV 相关抗原的表达,揭示 HCV 在 HCC 发生中的可能作用。方法应用免疫组化 SP 法定位46例 HCC 组织及38例癌旁组织中 HCV 核心、NS_3、NS_5抗原和 HBsAg。结果癌组织中各抗原的检出率分别为21.7%、21.7%、15.2%和19.6%,癌旁组织中的检出率分别为36.8%、34.2%、31.6%和78.9%。阳性染色细胞呈弥漫、灶状和散在分布.各抗原主要位于肝细胞和肝癌细胞的胞浆内,一些 NS_3、NS_5抗原可位于肝细胞的细胞膜上。阳性染色信号呈细颗粒状,少量的核心和 NS_5抗原可呈均质状.癌旁组织核心抗原阳性表达区可见淋巴细胞聚集,未发现 NS_3和 NS_5抗原表达与病理改变之间的联系。结论 HCV 可能是 HCC 的另一重要致病因素。
Objective To investigate the expression of HCV-associated antigen in liver tissue of patients with primary hepatocellular carcinoma (HCC) and to reveal the possible role of HCV in the development of HCC. Methods Immunohistochemical SP method was used to locate HCV core, NS_3, NS_5 antigen and HBsAg in 46 HCC tissues and 38 adjacent tissues. Results The detection rates of each antigen in cancer tissues were 21.7%, 21.7%, 15.2%, and 19.6%, respectively. The detection rates in adjacent tissues were 36.8%, 34.2%, 31.6%, and 78.9%, respectively. Positively stained cells showed diffuse, focal and scattered distributions. The antigens were mainly located in the cytoplasm of hepatocytes and hepatoma cells. Some NS3 and NS5 antigens were located on the cell membrane of hepatocytes. The positive staining signal was fine-grained, a small amount of core and NS_5 antigen could be homogenous. Lymphocyte aggregation was observed in the para-cancerous tissue core antigen positive expression area, and no correlation was found between the expression of NS_3 and NS_5 antigen and pathological changes. Conclusion HCV may be another important pathogenic factor of HCC.
