目的:以层次分析法结合量效对比法优选荆芥总黄酮的提取工艺条件。方法:选取料液比、乙醇体积分数、提取时间及提取次数为考察因素,以人克隆结肠腺癌细胞(Caco-2细胞)抑制率为指标,采用正交试验优选荆芥总黄酮的最佳提取工艺,并通过SPSS 19.0软件对总黄酮含量、橙皮苷含量及出膏率与药效学指标进行Pearson相关系数分析,确定量效对比指标,进而采用层次分析法确定总黄酮含量与橙皮苷含量权重系数范围。结果:最佳提取工艺条件为:15倍量75%乙醇,回流提取3次,每次2 h;总黄酮含量与橙皮苷含量的权重系数范围分别为0.7500~0.9000、0.1000~0.2500。结论:以基于AHP法确定的总黄酮含量、橙皮苷含量权重系数范围的综合评分代替抑制率作为优选最佳提取工艺的指标是科学合理的,且该优选工艺简便、实用,为中药有效成分的提取工艺筛选指标的确定提供新思路。
OBJECTIVE: To optimize the extraction conditions of total flavonoids from Nepeta by the analytic hierarchy process (AHP) and the quantitative comparison. Methods: The ratio of solid to liquid, the volume of ethanol, the extraction time and the number of extraction were selected as the investigation factors. Results: The optimal extraction conditions were as follows: 15 times 75% ethanol, reflux extraction 3 times, each time 2 h; weight coefficient of total flavonoids and hesperidin content were 0.7500 ~ 0.9000, 0.1000 ~ 0.2500. Conclusion: It is scientific and reasonable to use comprehensive score of hesperidin content weight coefficient to determine the optimal total flavonoid content and hesperidin content weight coefficient range as index of optimal and optimal extraction process, and the optimal process is simple and practical, which is the effective component of traditional Chinese medicine The extraction process screening indicators to determine the new ideas.
