目的观察白藜三醇(Res)对人外周血内皮祖细胞(EPCs)功能的影响。方法密度梯度离心法获取人外周血单个核细胞,培养7d后,收集贴壁细胞并加入Res(1、10、25、50μmol/L)干预一定时间(6、12、24、48h)。激光共聚焦显微镜和流式细胞术鉴定EPCs,分别观察EPCs的增殖、黏附、迁移和体外血管生成能力。结果Res促进外周血EPCs扩增,50μmol/LRes作用24h对EPCs数量的影响最为显著(P<0.01)。Res也显著改善了外周血EPCs的增殖、黏附、迁移和体外血管生成能力。结论Res可增加EPCs数量并改善其功能。
Objective To observe the effect of Res on the function of human peripheral blood endothelial progenitor cells (EPCs). Methods Peripheral blood mononuclear cells were obtained by density gradient centrifugation. After cultured for 7 days, adherent cells were collected and treated with Res (1, 10, 25, 50 μmol/L) for a certain period of time (6, 12, 24, 48 h). EPCs were identified by laser confocal microscopy and flow cytometry. The proliferation, adhesion, migration, and in vitro angiogenic capacity of EPCs were observed. Results Res promoted the amplification of EPCs in peripheral blood, and the effect of 50μmol/LRes for 24h had the most significant effect on the number of EPCs (P<0.01). Res also significantly improved peripheral blood EPCs proliferation, adhesion, migration, and in vitro angiogenesis. Conclusion Res can increase the number of EPCs and improve their function.
