目的 :提取正常人IL 10基因。方法 :利用RT PCR方法 ,自行设计并合成一对引物。结果 :成功地从活化的正常人外周血单个核细胞扩增出hIL 10cDNA ,并定向克隆入pUC18中 ,经过序列测定证实 ,为IL 10成熟肽cDNA基因。结论 :IL 10基因的获得 ,为进一步在大肠杆菌中高效表达有活性的重组hIL 10 ,更深入地研究其作用机理 ,以及临床应用奠定了基础。
Objective: To extract normal human IL 10 gene. Methods: A pair of primers was designed and synthesized by RT-PCR. RESULTS: hIL 10 cDNA was successfully amplified from activated normal human peripheral blood mononuclear cells and cloned into pUC18. The sequence was used to identify the IL 10 mature peptide cDNA. Conclusion: The obtainment of IL 10 gene provides a basis for further study of its mechanism of action and clinical application in order to further express highly active recombinant hIL 10 in E. coli.
