为研究CD137在正常人静止状态和活化T细胞上的表达特点及CD137单抗协同PHA 刺激T细胞增殖作用。利用流式细胞术测定CD137的表达,利用3H-TdR掺入法测定T细胞增殖。结果显示:静止T细胞上CD137表达率为(0.65±0.05)% ,经PHA 刺激后24、48、72h,T 细胞上CD137表达率分别为(8.16±1.28)% 、(12.39±2.15)% 、(21.6±4.36)% 。CD+4 T细胞和CD+8 T细胞均可表达CD137,且CD+4 T细胞的表达率高于CD+8 T细胞。单独用CD137单抗对T细胞的刺激指数(SI)为1.04,单独用PHA 刺激指数为2.31,不同浓度CD137单抗(1、5、10)m g/L和PHA 共同作用刺激指数分别为2.83、4.25、5.74。CD137McAb(10m g/L)+ PHA 组刺激指数比其他各组均明显增高(P< 0.05),并随CD137McAb 浓度增高对T 细胞的增殖逐渐增高。表明静止T细胞上不表达CD137,T细胞活化后,CD137表达量逐渐增加。CD137单抗可显著增加PHA对T细胞的增殖作用,而在无PHA 时,CD137单抗对T细胞无增殖作用
To study the expression of CD137 on quiescent human normal and activated T cells, CD137 monoclonal antibody synergizes with PHA to stimulate T cell proliferation. CD137 expression was measured by flow cytometry and T cell proliferation was measured by 3H-TdR incorporation. The results showed that the expression rate of CD137 on resting T cells was (0.65 ± 0.05)%, the positive rate of CD137 on T cells was (8.16 ± 1.28)% at 24, 48 and 72 h after PHA stimulation, , (12.39 ± 2.15)%, (21.6 ± 4.36)%. CD137 was expressed on both CD + 4 T cells and CD + 8 T cells, and the expression of CD + 4 T cells was higher than that of CD + 8 T cells. The stimulation index (SI) of T cells by CD137 alone was 1.04, the stimulation index by PHA alone was 2.31, and stimulation with different concentration of CD137 monoclonal antibody (1,5,10) m g / L and PHA The indices were 2.83, 4.25 and 5.74 respectively. The stimulation index of CD137McAb (10m g / L) + PHA group was significantly higher than that of other groups (P <0.05), and the proliferation of T cells was gradually increased with the increase of CD137McAb concentration. CD137 showed no expression of resting T cells, T cell activation, CD137 expression gradually increased. CD137 mAb significantly increased the proliferative effect of PHA on T cells, while in the absence of PHA, CD137 mAb did not proliferate on T cells
