目的:观察黄芩素(HQS)对冠脉结扎所致的大鼠缺血心肌的保护作用。方法:将60只大鼠随机分为假手术组10只及造模组50只,造模组大鼠麻醉开胸后结扎冠状动脉左前降支复制急性心肌梗死模型。手术后将造模成功的大鼠随机分为5组,假手术组和模型组静脉注射氯化钠注射液5 mL/kg,阳性对照组静脉注射硝酸甘油(nitroglyccrin,NG)0.3 mg/kg,受试药高、中、低剂量组分别给HQS 160 mg/kg、80 mg/kg、40 mg/kg。于冠脉结扎前、结扎后及给药后不同时间点监测心电图,标测ST段和T波(ST—T);于冠脉结扎6 h后行心肌氯化硝基四氮唑蓝(NBT)染色,测定心肌梗死重量。结果:不同剂量HQS可不同程度地降低S点位移、减轻心肌梗死重量。结论:黄芩素对冠脉结扎致大鼠急性心肌缺血损伤具有良好的保护作用,为中药抗心肌缺血损伤提供思路。
Objective: To observe the protective effect of baicalein (HQS) on rat ischemic myocardium induced by coronary artery ligation. METHODS: Sixty rats were randomly divided into 10 sham-operated groups and 50 model-building groups. Rats in the modeling group were anesthetized and opened and left anterior descending coronary artery was ligated to replicate the acute myocardial infarction model. Rats with successful modeling were randomly divided into 5 groups after surgery. Sham operation group and model group were intravenously injected with 5 mL/kg sodium chloride injection, and positive control group was intravenously injected with 0.3 mg/kg of nitroglycrine (NG). HQS 160 mg/kg, 80 mg/kg, and 40 mg/kg were given to the high, medium, and low dose groups of the test drug. Electrocardiogram was monitored before coronary artery ligation, after ligation, and at different time points after administration. ST segment and T wave (ST-T) were imaged; myocardial nitroblue tetrazolium blue (NBT) was established 6 h after coronary artery ligation. ) staining to determine the weight of myocardial infarction. Results: Different doses of HQS can reduce S point displacement and reduce myocardial infarction weight. Conclusion: Baicalein has a good protective effect on acute myocardial ischemic injury induced by coronary artery ligation in rats, and provides a guide for traditional Chinese medicine to resist myocardial ischemic injury.
