目的:探讨人卵巢癌细胞A2780中的HES1表达与人卵巢癌侵袭及转移的关系。方法:分别运用实时RT-PCR、WesternBlotting及细胞免疫荧光法检测γ分泌酶抑制剂作用前后A2780细胞中HES1的表达水平,Transwell小室试验及MTT法分别用于检测细胞的侵袭、迁移及黏附能力。结果:下调A2780细胞中HES1的表达水平后,A2780细胞的黏附、侵袭及转移能力下降,差异有统计学意义(P<0.01)。结论:HES1的表达与人卵巢癌细胞的侵袭及转移密切相关,下调其表达可抑制卵巢癌细胞的黏附、侵袭及转移能力。
Objective: To investigate the relationship between HES1 expression and invasion and metastasis in human ovarian cancer cell line A2780. Methods: The expression of HES1 in A2780 cells was detected by real-time RT-PCR, Western Blotting and immunofluorescence assay respectively. Transwell chamber assay and MTT assay were used to detect the invasion, migration and adhesion of A2780 cells. Results: After HES1 was down-regulated in A2780 cells, the adhesion, invasion and metastasis of A2780 cells were decreased, with statistical significance (P <0.01). Conclusion: The expression of HES1 is closely related to the invasion and metastasis of human ovarian cancer cells. Downregulating the expression of HES1 can inhibit the adhesion, invasion and metastasis of ovarian cancer cells.
