目的:探讨局部应用睫状神经营养因子(CNTF)对成年大鼠面神经损伤的修复作用。方法:将雄性SD大鼠颞骨内面神经垂直段横断后端端吻合,CNTF组以CNTF明胶海绵覆盖损伤处;对照组以生理盐水(SAL)的明胶海绵覆盖损伤处。术后CNTF组每周2次耳后皮下注射CNTF5μg,对照组同法注射等量SAL。术后2,4和12周各组进行电生理学、组织病理学观察及形态学定量分析。结果:术后2周,两组电刺激面神经均未能引发面肌兴奋,有髓纤维计数t检验,两组差异有显著性意义(P<0.05)。术后4周面神经复合肌动作电位(CMAP)CNTF组潜伏期明显短于对照组,有髓轴突计数CNTF组多于对照组(P<0.05)。术后12周,CMAP潜伏期、有髓纤维计数,两组差异均无显著性意义(P>0.05)。结论:局部应用CNTF可促进成年大鼠面神经损伤的修复。
Objective: To investigate the effect of topical application of ciliary neurotrophic factor (CNTF) on facial nerve injury in adult rats. Methods: The vertical sections of the medial facial nerve in the temporal bone of male Sprague-Dawley rats were anastomosed transversely, the CNTF group was covered with CNTF gelatin sponge, and the control group was covered with saline gelatin sponge. After operation, CNTF was injected subcutaneously twice a week twice a week into the CNTF group, while the control group received the same dose of SAL. Electrophysiology, histopathological observation and morphometric analysis were performed on the 2, 4 and 12 weeks after operation. Results: After 2 weeks of operation, facial nerve stimulation of both groups failed to induce facial muscle excitement and myelinated fiber count t test. There was significant difference between the two groups (P <0.05). The latent period of facial nerve complex muscle action potential (CMAP) CNTF group was significantly shorter than that of control group at 4 weeks after operation. The number of myelinated axon count CNTF group was more than that of control group (P <0.05). There was no significant difference between the two groups in the CMAP latency and the number of myelinated fibers 12 weeks after operation (P> 0.05). Conclusion: Topical application of CNTF can promote the repair of facial nerve injury in adult rats.
