目的研究奥曲肽单独及与转化生长因子-α(TGF-α)联用对人肝癌细胞增殖的影响,并探讨其机制。方法细胞增殖变化采用MTT比色法、绘制细胞生长曲线及流式细胞术(FCM)检测,细胞中TGF-α的表达采用放射免疫法及逆转录-聚合酶链反应(RT-PCR)检测。结果奥曲肽能抑制肝癌细胞增殖,且其抑制作用呈浓度和时间依赖性。奥曲肽(1μg/ml)作用48h,对TGFα(1μg/L、5μg/L)刺激的人肝癌细胞增殖抑制作用明显。两者同时作用于细胞较TGF-α单独作用,细胞生长曲线下移,增殖指数(PI)值明显降低(P<0.01)。加入奥曲肽后,上清液中TGF-α含量在相应时间点均较对照组明显减少,抑制率为15.02%~26.71%;TGF-αmRNA指数较对照组降低。结论奥曲肽能抑制人肝癌细胞增殖,且对TGF-α刺激的人肝癌细胞增殖也有抑制作用,其机制可能与奥曲肽抑制肝癌细胞TGF-α分泌有关。
Objective To study the effects of octreotide alone and combined with transforming growth factor-α (TGF-α) on the proliferation of human hepatocellular carcinoma cells and its mechanism. Methods Cell proliferation was measured by MTT assay. Cell growth curve and flow cytometry (FCM) were used to detect cell proliferation. The expression of TGF-α in the cells was detected by radioimmunoassay and reverse transcription-polymerase chain reaction (RT-PCR). The results octreotide can inhibit the proliferation of hepatocellular carcinoma cells, and its inhibitory effect in a concentration and time-dependent manner. Octreotide (1μg / ml) for 48h, inhibit the proliferation of human hepatoma cells stimulated by TGFα (1μg / L, 5μg / L) obviously. The effect of both of them on the cells was lower than that of TGF-α alone, and the cell growth curve shifted down, and the proliferation index (PI) value decreased significantly (P <0.01). After addition of octreotide, the content of TGF-α in the supernatant decreased significantly at the corresponding time points compared with the control group, with the inhibition rate of 15.02% -26.71%. The TGF-αmRNA index was lower than that of the control group. Conclusion Octreotide can inhibit the proliferation of human hepatocellular carcinoma cells and inhibit the proliferation of human hepatoma cells stimulated by TGF-α, which may be related to the inhibition of the secretion of TGF-α by octreotide.
