目的 探讨云南汉族系统性红斑狼疮 (SLE)患者抗SSA和SSB抗体与HLA DRB1、DQA1、DQB1等位基因及单体型的相关性。方法 采用多聚酶链反应 序列特异性引物 (PCR SSP)技术对 6 3例云南汉族SLE患者和 5 4名同民族健康对照进行DRB1、DQA1、DQB1基因分型。结果 云南汉族SLE患者中DR15 (P <0 0 1)、DR16 (P <0 0 5 )、DQA1 0 10 2 (P <0 0 5 )、DQA1 0 10 3(P<0 0 1)、DQB1 0 6 0 1(P <0 0 5 )等位基因频率明显增高 ;抗SSA和SSB抗体阳性的SLE病人中DQA1 0 10 3频率均显著增高 (P =0 0 42 ,P =0 0 0 6 ) ;抗SSB抗体阳性的SLE患者中DQA1 0 5 0 1频率均显著增高 (P =0 0 0 9)。结论 云南汉族SLE抗SSA和SSB抗体的产生与DQA1 0 10 3等位基因相关 ;抗SSB抗体的产生还与DQA1 0 5 0 1相关。
Objective To investigate the association of anti-SSA and SSB antibodies with HLA DRB1, DQA1 and DQB1 alleles and haplotypes in patients with systemic lupus erythematosus (SLE) in Yunnan Province. Methods Genotyping of DRB1, DQA1 and DQB1 was performed on 63 SLE patients in Yunnan Han nationality and 54 healthy controls of the same ethnic origin by polymerase chain reaction sequence specific primers (PCR SSP). Results There was no significant difference in DR15 (P <0.01), DR16 (P <0 05), DQA1 0 102 (P 0 05), DQA 1 0 10 3 (P 0 01), DQB 1 0 The frequency of DQA1 0 10 3 was significantly higher in SLE patients with SSA and SSB antibodies (P = 0.0042, P = 0.0060). The frequency of DQA1 0 5 0 1 was significantly increased in SLE patients with anti-SSB antibodies (P = 0 0 0 9). Conclusions The generation of anti-SSA and SSB antibodies in Yunnan Han population is related to the DQA1 0 10 3 allele. The production of anti-SSB antibodies is also associated with DQA1 0 5 0 1.
