目的鲍氏不动杆菌(ABA)医院感染株亲缘性3种分析方法学的比较。方法分别采用脉冲场凝胶电泳(PFGE),扩增片段长度多态性分析(AFLP)和多基因聚类分析对27株鲍氏不动杆菌临床分离株进行分析。结果PFGE分析显示其中的19株鲍氏不动杆菌为同一克隆传播;AFLP显示24株鲍氏不动杆菌为同一克隆传播;而多基因聚类分析则显示,携带8种基因(TEM、OXA-23群、ADC、aac(3)-Ⅰ、aac(6′)-Ⅰ、ant(3″)-Ⅰ、qacE△1-sul1i、ntⅠ1)、携带7种基因(TEM、ADC、aac(3)-Ⅰ、aac(6′)-Ⅰ、ant(3″)-Ⅰ、qacE△1-sul1i、ntⅠ1)、携带6种基因(TEM、ADC、aac(3)-Ⅰ、ant(3″)-Ⅰ、qacE△1-sul1i、ntⅠ1)的3种ABA克隆为医院的流行株(3个克隆)。结论鲍氏不动杆菌医院感染株亲缘性分析中多基因聚类分析法分辨率高于PFGE与AFLP法。
Objective To compare the three methods of analyzing the relatives of hospital infection with Acinetobacter baumannii (ABA). Methods The clinical isolates of 27 strains of Acinetobacter baumannii were analyzed by pulsed-field gel electrophoresis (PFGE), amplified fragment length polymorphism (AFLP) and polygene cluster analysis respectively. Results PFGE analysis showed that 19 strains of Acinetobacter baumannii were transmitted by the same clone. AFLP showed that 24 strains of Acinetobacter baumannii were transmitted by the same clone. However, multiple gene cluster analysis showed that 8 strains of genes carrying TEM, OXA- ADC, aac (3) -Ⅰ, aac (6 ’) - Ⅰ, ant (3 “) - Ⅰ, qacE △ 1-sul1i, ntⅠ1) AAC (3) - Ⅰ, ant (3 ”) - Ⅰ, qacE △ 1 - sul1i, nt Ⅰ 1) Ⅰ, qacE △ 1-sul1i, ntⅠ1) were identified as epidemic strains (3 clones) in the hospital.Conclusion The multi-gene clustering analysis of the relatives of Acinetobacter baumannii isolates was more effective than PFGE With AFLP method.
