为制备抗马来丝虫感染期幼虫表膜抗原(S)、排泄分泌抗原(ES_m),以及马颈盘尾丝虫微丝蚴排泄分泌抗原(ES_c)的单克隆抗体,作者介绍了一种脾内注射活虫体的快捷免疫方法。脾内抗原注射后3~4天进行细胞融合。较之传统方法省时,免疫所用虫量亦少。 BALB/c ByJ小鼠(6~8周♀),用乙醚麻醉,于腹部脾区作一长2cm切口,暴露脾尖。为使抗原在脾内均匀分布,25号针纵向刺入脾一定深度,边退针边注入含活虫体的无血清培养液50μl,3~4天后取脾。
