目的 研究三氧化二砷 (As2 O3)对人胃癌细胞CD4 4、p5 3、nm2 3、H ras及PCNA等五种转移相关基因的影响 ,探讨其抗肿瘤作用。方法 以As2 O3作用于体外培养的人胃癌细胞株 ,通过免疫组化技术对以上五种基因编码蛋白的表达进行检测。结果 经As2 O3作用的人胃癌细胞CD4 4、p5 3、PCNA基因编码蛋白表达减少 ,nm2 3基因编码蛋白表达改变不明显 ,H ras基因编码蛋白表达相对处于较低水平。结论 初步证实 ,As2 O3的抗肿瘤作用可能与下调CD4 4、p5 3、PCNA基因编码蛋白表达有关 ,从而抑制肿瘤的转移。
Objective To study the effects of arsenic trioxide (As2O3) on five metastasis-related genes including human gastric cancer CD4 4, p53, nm23, H ras, and PCNA, and to explore their anti-tumor effects. Methods As2O3 was used to culture human gastric cancer cell lines in vitro. The expression of the above five genes was detected by immunohistochemistry. Results The protein expression of CD4 4, p53, and PCNA genes was decreased in As2O3-treated human gastric cancer cells, and the protein expression of nm23 gene was not changed significantly. The protein expression of H ras gene was relatively low. Conclusion It is initially confirmed that the antitumor effect of As2O3 may be related to the down-regulation of CD44, p53, and PCNA gene-encoded protein expression, thereby inhibiting tumor metastasis.
