目的 :探讨雷公藤红素对鼻息肉肥大细胞凋亡的诱导作用及机制。方法 :雷公藤红素 (2μm ol/ L )与鼻息肉组织块在体外共育 ,应用 TUNEL 标记检测凋亡细胞情况 ,免疫组化检测凋亡相关蛋白的表达。结果 :未加药对照组 6和 12 h(n=10 ) 10个相邻高倍视野 TU NEL 阳性细胞数培养分别为 (7.8± 7.7)和 (10 .6± 4.5 )个 ,雷公藤红素组 (n=10 )分别为 (6 2 .3±42 .7)和 (76 .3± 39.3)个 ,二者相比有显著性差异 (P<0 .0 5 )。甲胺蓝染色明确显示肥大细胞呈凋亡形态改变 ,包括核浓缩、凋亡小体形成等 ,少见上皮细胞和成纤维细胞呈凋亡形态改变。雷公藤红素组促凋亡相关蛋白 Fas、C- myc、Bax表达增加 ,抑凋亡相关蛋白 bcl- 2表达下降。 结论 :雷公藤红素能诱导鼻息肉肥大细胞凋亡 ,凋亡的发生与其上调促凋亡基因的表达和下调抑凋亡基因的表达有关。
Objective : To investigate the induction and mechanism of apoptosis induced by tripterine on nasal mast cells. METHODS: Tripterine (2 μmol/L) was incubated with nasal polyp tissue blocks in vitro. TUNEL labeling was used to detect apoptotic cells. Immunohistochemistry was used to detect the expression of apoptosis-related proteins. RESULTS: The number of TU NEL-positive cells cultured in 10 adjacent high-power fields at 6 and 12 h (n=10) in the undosed control group was (7.8 ± 7.7) and (10 .6 ± 4.5), respectively. Tripterine group (n=10) were (62.3 ± 42.7) and (76.3 ± 39.3), respectively, and there was a significant difference between the two (P < 0.05). Methylamine blue staining clearly showed apoptotic morphology changes in mast cells, including nuclear condensation, formation of apoptotic bodies, and rare epithelial cells and fibroblasts showed apoptotic morphology changes. The expression of apoptosis-associated proteins Fas, C-myc and Bax was increased in the tripterine group, and the expression of the apoptosis-inhibiting protein bcl-2 was decreased. Conclusion: Tripterine can induce apoptosis of mast cells in nasal polyps. The occurrence of apoptosis is related to up-regulation of pro-apoptotic gene expression and down-regulation of anti-apoptotic gene expression.
