目的对分离自外阴阴道念珠菌病患者阴道的念珠菌进行基因分型,并对不同菌株间的亲缘关系作相似性分析。方法用形态学及生化方法对分离到的致病性念珠菌进行鉴定,然后采用随机扩增多态性DNA技术(RAPD)对45株临床分离株基因组DNA进行扩增,并将DNA扩增带型进行聚类分析。结果30株白念株菌可以分成7群共19个亚群,15株非白念珠菌(包括光滑念珠菌6株,克鲁斯念珠菌、近平滑念珠菌和热带念珠菌各3株)至少可鉴定到种水平。白念珠菌种内不同菌株间的相似性系数在90%以上,不同念珠菌种间的相似性系数介于80%~90%。结论外阴阴道念珠菌病的主要致病菌是白念珠菌,用RAPD技术可将之分成不同的基因组型别。
Objective To genotype Candida from the vagina of patients with vulvovaginal candidiasis and to analyze the similarities among different strains. Methods Morphological and biochemical methods were used to identify the pathogenic Candida isolates. Genomic DNA of 45 clinical isolates was amplified by random amplified polymorphic DNA (RAPD) and the DNA was amplified Type cluster analysis. Results Thirty strains of Candida albicans were divided into 7 subgroups and 19 subgroups, at least 15 non-Candida albicans (including 6 Candida glabrata, 3 Candida krusei, 3 Candida parapsilosis and 3 Candida tropicalis) Can be identified to species level. Candida albicans within the different strains of the similarity coefficient of more than 90%, different Candida species similarity coefficient between 80% to 90%. Conclusion The main pathogen of vulvovaginal candidiasis is Candida albicans, which can be divided into different genotypes by RAPD technique.
