目的探究肿瘤相关巨噬细胞促进肝癌细胞转移的潜在分子机制。方法巨噬细胞经刺激后与肝癌细胞共培养,利用RT-PCR和流式细胞术检测肿瘤相关巨噬细胞对肝癌细胞c-Met分子表达的影响,利用Transwell细胞迁移实验检测HGF-c-Met通路对肝癌细胞迁移能力的影响。结果 THP-1来源的巨噬细胞与肝癌细胞共培养后,肝癌细胞c-Met分子在mRNA和蛋白水平的表达均有增强;与巨噬细胞共培养的肝癌细胞,在重组蛋白HGF诱导作用下,其迁移能力明显增强。结论肿瘤相关巨细胞可以上调肝癌细胞c-Met分子的表达,高表达的c-Met分子增强了肝癌细胞的迁移能力。
Objective To investigate the potential molecular mechanism of tumor-associated macrophages in promoting the metastasis of hepatocellular carcinoma cells. Methods Macrophages were co-cultured with hepatocellular carcinoma cells after stimulation. The effect of tumor-associated macrophages on the expression of c-Met was detected by RT-PCR and flow cytometry. Transwell migration assay was used to detect the expression of HGF-c-Met Pathway on the migration of hepatocellular carcinoma cells. Results The co-culture of THP-1-derived macrophages with hepatocellular carcinoma cells enhanced the expression of c-Met in hepatocellular carcinoma cells at mRNA and protein level. The co-cultured hepatoma cells with THP-1 cells under the induction of recombinant protein HGF , Its migration ability is obviously enhanced. Conclusion Tumor-associated giant cells can up-regulate the expression of c-Met in hepatocellular carcinoma cells. High expression of c-Met enhances the migration of hepatoma cells.
