目的探讨外周血miR-29a-3p、miR-365a-3p在活动性肺结核和结核潜伏感染者中的差异表达。方法收集深圳市南山区慢性病防治院结核病防治科门诊2012年1~10月诊治的活动性肺结核患者30例,及同期发现的结核潜伏感染者30例。采集全血,提取总RNA,利用定量PCR方法检测外周血miR-29a-3p、miR-365a-3p的表达。结果结核潜伏感染者外周血中miR-29a-3p(1.16±0.65)明显高于活动性肺结核患者的表达(0.71±0.32),差异有统计学意义(P<0.05);外周血miR-365a-3p的表达在结核潜伏感染者(0.43±0.51)与活动性肺结核患者(0.32±0.71)之间差异无统计学意义(P>0.05);进一步分析发现,结核潜伏感染者外周血miR-29a-3p表达与结核菌特异性酶联免疫斑点数呈负相关(r=-0.42,P=0.02)。结论 miR-29a-3p在肺结核感染、发病中可能起到重要作用,可能成为诊断结核潜伏感染者的标志物。
Objective To investigate the differential expression of miR-29a-3p and miR-365a-3p in peripheral blood of active tuberculosis and tuberculosis latent infection. Methods Thirty cases of active pulmonary tuberculosis (TB) diagnosed and treated from January to October in 2012 in Nanshan District, Shenzhen were enrolled in this study. Thirty patients with tuberculosis latent infection were detected in the same period. Whole blood was collected and total RNA was extracted. The expression of miR-29a-3p and miR-365a-3p in peripheral blood was detected by quantitative PCR. Results The expression of miR-29a-3p (1.16 ± 0.65) in peripheral blood of patients with tuberculosis latent infection was significantly higher than that of patients with active tuberculosis (0.71 ± 0.32), the difference was statistically significant (P <0.05) There was no significant difference in the expression of 3p between TB latent infection (0.43 ± 0.51) and active pulmonary tuberculosis (0.32 ± 0.71). Further analysis showed that miR-29a- 3p expression was negatively correlated with TB-specific ELISP (r = -0.42, P = 0.02). Conclusion miR-29a-3p may play an important role in the pathogenesis of pulmonary tuberculosis infection and may be the marker of tuberculosis latent infection.
