目的观察茶碱对嗜酸细胞(Eos)凋亡的影响。方法采取15例正常人外周血,体外用茶碱(10-4M)和rhIL-5(10-8M)预处理,以地塞米松(10-4M)为对照,用反转录-聚合酶链式反应(RT-PCR)和流式细胞仪法分别从mRNA和蛋白水平测定Eosbcl-2和bax的表达,以评价茶碱对Eos凋亡的影响。结果未受刺激的Eos正常表达Bax,而Bcl-2蛋白和mRNA则无法测得;IL-5能够上调bcl-2mRNA的转录(P<0.01)和Bcl-2蛋白的表达(P<0.01),这一作用被茶碱和地塞米松所抑制;IL-5对baxmRNA的转录和Bax蛋白的表达则无影响(P>0.05),茶碱和地塞米松对baxmRNA的转录和Bax蛋白的表达亦无影响。且茶碱和地塞米松只对bcl-2表达的抑制有协同作用。结论茶碱对Eos凋亡的影响可能由于干预了Bcl-2的表达。
Objective To observe the effect of theophylline on the apoptosis of eosinophils (Eos). Methods Peripheral blood samples from 15 normal subjects were pretreated with theophylline (10-4M) and rhIL-5 (10-8M) in vitro. Dexamethasone (10-4M) The expression of Eosbcl-2 and bax were detected by RT-PCR and flow cytometry respectively at mRNA and protein levels to evaluate the effect of theophylline on Eos apoptosis. Results The unstimulated Eos normally expressed Bax, while the Bcl-2 protein and mRNA could not be measured. IL-5 upregulated the expression of bcl-2 mRNA and Bcl-2 protein (P <0.01) This effect was inhibited by theophylline and dexamethasone; IL-5 had no effect on the transcription of bax mRNA and the expression of Bax protein (P> 0.05). The theophylline and dexamethasone on bax mRNA transcription and Bax protein expression no effect. Theophylline and dexamethasone only synergistically inhibit the expression of bcl-2. Conclusion Theophylline may affect the apoptosis of Eos probably by interfering with the expression of Bcl-2.
