目的探讨早孕期蜕膜组织细胞凋亡异常与妊娠维持的关系。方法应用TUNEL法检测凋亡的发生,原位杂交和免疫组化法检测Fas、FasL的表达。结果(1)正常早孕41d蜕膜组织细胞大量凋亡,Fas、FasL的mRNA及蛋白表达较强;(2)正常早孕54d凋亡细胞明显减少,Fas、FasL的表达明显减弱;(3)早孕55d自然流产组,蜕膜组织大量凋亡,且Fas、FasL的表达较正常早孕54d显著增强(P<0.05)。结论早孕期蜕膜组织凋亡异常可能是自然流产的机制之一,Fas途径可能是诱导异常凋亡的重要因素。
Objective To investigate the relationship between abnormal apoptosis of decidual cells in early pregnancy and maintenance of pregnancy. Methods The occurrence of apoptosis was detected by TUNEL method. The expressions of Fas and FasL were detected by in situ hybridization and immunohistochemistry. Results (1) A large number of apoptotic cells were found in the decidua of normal pregnant women on the 41th day of pregnancy, and the mRNA and protein expressions of Fas and FasL were strong. (2) The apoptotic cells were significantly decreased at 54th day of pregnancy and the expression of Fas and FasL was significantly weakened. (3) In spontaneous abortion group and decidua group, a large number of apoptosis occurred on day 55d, and the expression of Fas and FasL increased significantly (P <0.05) 54 d after normal pregnancy. Conclusion Abnormal apoptosis of decidua in early pregnancy may be one of the mechanisms of spontaneous abortion. Fas pathway may be an important factor in inducing abnormal apoptosis.
