目的:观察三氧化二砷(As2O3)对肝癌HepG2细胞体外黏附、迁移、侵袭和增殖的抑制作用。方法:采用四甲基偶氮唑盐(MTT)法观察As2O3对HepG2细胞黏附能力、增殖能力的影响,以Transwell检测As2O3对HepG2细胞迁移、侵袭能力的影响。结果:As2O3作用后30,60,90,120min时的细胞黏附率较对照组均明显下降(P<0.05);As2O3作用后游走及侵袭穿膜细胞数也较对照组明显下降(P<0.01);不同浓度的As2O3均可显著抑制HepG2细胞的生长,抑制作用在一定范围内呈浓度及时间依赖性(P<0.01)。结论:As2O3可明显抑制HepG2细胞细胞黏附、迁移、侵袭和增殖的能力。
Objective: To observe the inhibitory effect of arsenic trioxide (As2O3) on the adhesion, migration, invasion and proliferation of HepG2 cells in vitro. Methods: The effects of As2O3 on the adhesion and proliferation of HepG2 cells were observed by MTT method. The effects of As2O3 on the migration and invasion of HepG2 cells were detected by Transwell assay. RESULTS: The cell adhesion rates at 30, 60, 90 and 120 min after As2O3 treatment were significantly lower than those in control group (P <0.05) ; As2O3 can significantly inhibit the growth of HepG2 cells in a concentration-dependent and time-dependent manner (P <0.01). Conclusion: As2O3 can significantly inhibit HepG2 cell adhesion, migration, invasion and proliferation.
