目的分析脂肪来源干细胞(ADSCs)干预后辐射诱发胸腺损伤修复后的胸腺细胞周期蛋白D1、p53基因的表达。方法 C57BL/6小鼠随机分为:对照组、照射组、照射+ADSCs组和假照射+ADSCs组8只。照射组为辐射诱发胸腺淋巴瘤动物模型组,照射+ADSCs组为尾静脉注射EGFP质粒转染ADSCs。检测胸腺细胞凋亡和细胞周期。结果在ADSCs注射后第7天,照射组小鼠胸腺G1期细胞的百分比较对照组明显降低(P<0.05),而照射+ADSCs组小鼠胸腺G1期细胞的百分比较照射组明显增多;cyclinD1基因在ADSCs治疗组胸腺组织中的相对表达量较照射组增高,且差异有统计学意义(P<0.01);而p53基因在照射组胸腺组织中的相对表达量较ADSCs治疗组增高,且差异有统计学意义(P<0.05)。结论 ADSCs可能通过p53、cyclinD1等基因的表达水平变化来调控细胞周期,形成细胞G1期阻滞,抑制肿瘤的形成。
Objective To analyze the expressions of cyclin D1 and p53 genes in thymus after radiation - induced thymus injury after intervention of adipose derived stem cells (ADSCs). Methods C57BL / 6 mice were randomly divided into control group, irradiation group, irradiation + ADSCs group and sham irradiation + ADSCs group. The irradiation group was radiation-induced thymus lymphoma animal model group, irradiation + ADSCs group was injected EGFP plasmid into tail vein for ADSCs transfection. Detection of thymocyte apoptosis and cell cycle. Results At the 7th day after ADSCs injection, the percentage of G1 phase cells in the thymus of the irradiation group was significantly lower than that of the control group (P <0.05), while the percentages of the G1 phase cells in the irradiated + ADSCs group were significantly increased compared with the irradiated group. The expression of cyclinD1 The relative expression level of gene in thymus tissue of ADSCs group was higher than that of irradiation group, and the difference was statistically significant (P <0.01). The relative expression of p53 gene in thymus tissue of irradiation group was higher than that of ADSCs group There was statistical significance (P <0.05). Conclusion ADSCs may regulate the cell cycle through the expression of p53, cyclinD1 and other genes to form G1 cell cycle arrest and inhibit tumor formation.
