目的:探讨肝脂酶(hepatic lipase,HL)的基因-710T/C、-763A/G多态性在浙江金华地区不同年龄段男性人群中的分布及其与脂类代谢间的关系。方法:采用聚合酶链反应-限制性片段长度多态性(PCR-RFLP)检测浙江金华地区355名正常男性的肝脂酶基因-710T/C和-763A/G多态性,并探讨其基因多态性与血脂水平的关系。将355名研究对象按年龄分组,6~7岁组74名,18~19岁组109名,20~39岁组67名,40~73岁组105名。结果:HL-710T/C、-763A/G多态性的基因型和等位基因频率分布显示,40~73岁组与6~7岁组及18~19岁组间的差异均具有统计学意义(P<0.05)。HL-710T/C、-763A/G多态性与脂类代谢有关,HL-710T/C与-763A/G的连锁不平衡系数达0.788。结论:HL-710T/C与-763A/G间存在强连锁不平衡,HL-710T/C、-763A/G的多态性可能与脂质代谢有关。
Objective: To investigate the distribution of -710T / C and -763A / G polymorphisms of hepatic lipase (HL) gene in male population of different ages in Jinhua, Zhejiang Province and their relationship with lipid metabolism. Methods: Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to detect the polymorphisms of hepatic lipase gene-710T / C and -763A / G in 355 normal males in Jinhua, Zhejiang Province, Relationship between Polymorphism and Blood Lipid Level. A total of 355 subjects were grouped by age, 74 in the 6-7 age group, 109 in the 18-19 age group, 67 in the 20-39 age group and 105 in the 40-73 age group. Results: The frequency distribution of genotypes and alleles of HL-710T / C and -763A / G polymorphisms showed that the differences between 40-73 years old group and 6-7 years old group and 18-19 years old group were statistically significant Significance (P <0.05). HL-710T / C and -763A / G polymorphisms were related to lipid metabolism, and the linkage disequilibrium coefficient between HL-710T / C and -763A / G was 0.788. CONCLUSION: There is a strong linkage disequilibrium between HL-710T / C and -763A / G. The polymorphisms of HL-710T / C and -763A / G may be related to lipid metabolism.
