目的建立超高效液相色谱-串联质谱法(UPLC-MS/MS)研究淫羊藿素(ICT)在大鼠体内的组织分布。方法乙腈提取大鼠组织匀浆中ICT,在C18色谱柱上以含有甲酸和甲酸铵的乙腈-水二元溶剂梯度洗脱分离ICT和内标化合物香豆雌酚(CMT),负离子电喷雾选择反应监测方式定量分析ICT(m/z 367.1→297.1)和CMT(m/z 267.0→211.1)。采集大鼠单次腹腔注射10 mg/kg ICT后不同时间的心、肝、脾、肺、肾和肌肉组织并测定ICT浓度。结果 ICT在0.2~20 ng/mL浓度范围内线性关系良好,定量下限为0.2 ng/mL。ICT在肌肉中的半减期最长(12.46 h),在肝中有最高浓度(2949 ng/g)。结论 ICT广泛分布在各组织中,48 h各组织中ICT清除均超过93%,表明ICT在组织中无蓄积。
Objective To establish an ultra high performance liquid chromatography-tandem mass spectrometry (UPLC-MS / MS) system for studying the tissue distribution of icaritin (ICT) in rats. Methods The ICTs of rat tissue homogenates were extracted by acetonitrile. The ICT and internal standard compound coumestrol (CMT) were separated on a C18 column with acetonitrile - water binary solvent containing formic acid and ammonium formate. The negative ion electrospray ionization Response monitoring methods quantify ICT (m / z 367.1 → 297.1) and CMT (m / z 267.0 → 211.1). Heart, liver, spleen, lung, kidney and muscle tissue were collected at different time points after single intraperitoneal injection of 10 mg / kg ICT and the concentrations of ICT were measured. Results The linearity of ICT was good in the concentration range of 0.2 ~ 20 ng / mL with the lower limit of quantitation of 0.2 ng / mL. ICT has the longest half-life reduction in muscle (12.46 h) and the highest concentration in liver (2949 ng / g). Conclusion ICTs are widely distributed in various tissues, with ICT clearance over 93% at 48 hours indicating that there is no accumulation of ICT in the tissues.
