为优化牛蒡叶抗氧化活性物质的提取工艺,通过单因素试验考察了乙醇体积分数、提取时间、提取温度和液料比对提取物清除DPPH自由基活性能力的影响,在单因素试验的基础上,通过Box-Behnken中心组合试验设计对提取工艺进行优化,获得了牛蒡叶抗氧化活性物质提取条件的最佳工艺。结果表明:最佳提取工艺条件为乙醇体积分数58.6%、提取时间36.3min、提取温度52℃和液料比22mL/g,在此条件下牛蒡叶提取物对DPPH自由基清除率的最大理论值为97.40%。对此优化条件进行验证,重复3次,实测平均清除率为97.34%,表明该回归模型具有较好的预测性能,可用于指导生产实践。
In order to optimize the extraction process of antioxidant active substances from Arctium lappa L., the effect of ethanol volume fraction, extraction time, extraction temperature and liquid to material ratio on the DPPH radical scavenging activity was investigated by single factor experiments. Based on the single factor test , The optimum extraction process of Arctium lappa L. antioxidant active material was obtained through the combination of Box-Behnken central experimental design. The results showed that the optimum extraction conditions were as follows: ethanol volume fraction 58.6%, extraction time 36.3 min, extraction temperature 52 ℃ and liquid / solid ratio 22 mL / g. Under these conditions, the maximum theoretical value for the scavenging rate of DPPH 97.40%. The optimization conditions were verified and repeated 3 times, the measured average clearance rate was 97.34%, indicating that the regression model has good predictive performance and can be used to guide the production practice.
