目的探讨骨髓间充质干细胞(BMSCs)移植对大鼠脑缺血再灌注损伤海马区凋亡相关基因Livin和Caspase-3表达及神经元细胞凋亡的影响。方法实验动物分为假手术组、模型组和BMSCs组,进行神经功能评分,应用TTC法检测脑梗死体积,追踪PKH26标记的移植BMSCs,应用免疫组化方法和Western blot检测Livin、Caspase-3蛋白表达,应用TUNEL法检测细胞凋亡。结果 PKH26标记的BMSCs在海马区有表达。与模型组比较,BMSCs组神经功能评分显著降低(P<0.05),脑梗死体积显著减少(P<0.05)。BMSCs组与模型组相比Livin蛋白表达显著增高(P<0.05),Caspase-3蛋白表达明显下降(P<0.05),神经元细胞凋亡指数显著降低(P<0.05)。结论 BMSCs对脑缺血再灌注损伤具有保护作用,其作用机制可能与上调Livin表达,下调Caspase-3表达,抑制细胞凋亡有关。
Objective To investigate the effects of bone marrow mesenchymal stem cells (BMSCs) transplantation on the expression of Livin, Caspase-3 and neuronal apoptosis in hippocampus after cerebral ischemia-reperfusion injury in rats. Methods The experimental animals were divided into sham operation group, model group and BMSCs group. The neurological function was evaluated. The volume of cerebral infarction was detected by TTC method. The PKC26 labeled BMSCs were traced. The expressions of Livin and Caspase-3 protein were detected by immunohistochemistry and Western blot The apoptosis was detected by TUNEL method. Results PKH26-labeled BMSCs were expressed in the hippocampus. Compared with the model group, the neurological scores of BMSCs were significantly decreased (P <0.05), and the volume of cerebral infarction was significantly decreased (P <0.05). Compared with the model group, the expression of Livin protein in BMSCs group was significantly increased (P <0.05), Caspase-3 protein expression was significantly decreased (P <0.05), and apoptosis index in neurons was significantly decreased (P <0.05). Conclusion BMSCs have a protective effect on cerebral ischemia-reperfusion injury. The mechanism may be related to up-regulating Livin expression, down-regulating Caspase-3 expression and inhibiting apoptosis.
