目的探讨通解口服液对多器官功能障碍综合征(MODS)大鼠血清和回肠组织中IL-6、TNF-α的影响。方法将48只雄性SD大鼠随机分为空白组、模型组、阳性组、低剂量组、中剂量组和高剂量组,共6组,每组8只。各组均预先灌胃给予相应的干预措施,2天后,除空白组外,其余各组舌下静脉注射8μg/g脂多糖(LPS)复制MODS大鼠模型。观察大鼠生命体征(心电图、体温、呼吸幅度和频率)、血清指标(AST、ALT、CK、Cr)及血清和回肠组织IL-6、TNF-α含量。结果与空白组比较,模型组大鼠心率、体温、呼吸频率、血清指标(AST、ALT、CK、Cr)及血清和回肠组织TNF-α、IL-6含量差异有统计学意义(P<0.05,P<0.01)。与模型组比较,阳性组大鼠心率、体温差异有统计学意义(P<0.05);低、中、高剂量组大鼠心率差异有统计学意义(P<0.05,P<0.01)。与模型组比较,阳性组呼吸频率、血清指标及回肠组织中IL-6含量差异有统计学意义(P<0.05,P<0.01);高剂量组呼吸频率、血清指标、血清及回肠组织中TNF-α、IL-6含量差异有统计学意义(P<0.05,P<0.01);中剂量组血清指标、血清及回肠组织IL-6含量差异有统计学意义(P<0.05,P<0.01);低剂量组Cr水平差异有统计学意义(P<0.01)。与低剂量组比较,高剂量组血清ALT、AST、TNF-α、IL-6及回肠组织中IL-6含量,中剂量组的血清ALT含量差异有统计学意义(P<0.05,P<0.01)。与阳性组比较,低剂量组AST、ALT含量,高剂量组血清及回肠组织中IL-6含量差异有统计学意义(P<0.05,P<0.01)。结论通解口服液可有效降低脂多糖诱导的MODS大鼠血清及回肠组织中IL-6、TNF-α水平,且与其剂量具有相关性。
Objective To investigate the effects of Tongxie Oral Liquid on IL-6 and TNF-α in serum and ileum of multiple organ dysfunction syndrome (MODS) rats. Methods 48 male SD rats were randomly divided into blank group, model group, positive group, low dose group, medium dose group and high dose group, a total of 6 groups of 8 rats. The rats in each group were pre-gavaged with appropriate interventions. After 2 days, all the rats in each group were given sublingual ventricular injection of 8μg / g lipopolysaccharide (LPS) to replicate the model rats. The vital signs (electrocardiogram, body temperature, respiration rate and frequency), serum indexes (AST, ALT, CK, Cr) and IL-6 and TNF-α in serum and ileum were observed. Results Compared with the blank group, the heart rate, body temperature, respiratory rate, serum indexes (AST, ALT, CK, Cr) and the levels of TNF-α and IL-6 in the serum and ileum of the model group were significantly different , P <0.01). Compared with the model group, the difference of heart rate and body temperature in the positive group was statistically significant (P <0.05). The heart rate of the rats in the low, medium and high dose groups was significantly different (P <0.05, P <0.01). Compared with the model group, the positive rate of respiratory rate, serum index and ileal IL-6 content was significantly different (P <0.05, P <0.01); high dose group respiratory rate, serum indicators, serum and ileal TNF (P <0.05, P <0.01). Serum levels of IL-6 and IL-6 in serum and ileum in the middle dose group were significantly different (P <0.05, P <0.01) There was significant difference in Cr level between low-dose group and low-dose group (P <0.01). Compared with the low dose group, the levels of serum ALT, AST, TNF-α, IL-6, IL-6 in the ileum and the serum ALT in the middle dose group were significantly different (P <0.05, P <0.01 ). Compared with the positive group, the content of AST and ALT in the low dose group and the IL-6 content in the serum and ileum of the high dose group were significantly different (P <0.05, P <0.01). Conclusion Tong-Xie oral solution can effectively reduce the levels of IL-6 and TNF-α in serum and ileum of rats with MODS induced by lipopolysaccharide, and has a correlation with its dosage.
