高催乳素血症的妇女常伴有促性腺激素分泌功能的抑制和不孕,患者常发生闭经并缺乏雌激素的正反馈。虽已证实催乳素(PRL)对促性腺功能具抑制作用,但其抑制LH的作用机理尚不清楚。有证据提示高水平PRL通过增加下丘脑结节漏斗部多巴胺神经元的活动抑制LH分泌,也有人认为PRL对卵巢有直接抑制。本文试图通过垂体的离体培养观察PRL对垂体前叶的直接抑制作用。材料与方法为成熟雌性大鼠在切除卵巢二周后以断头法处死,取出垂体前叶,对半切开,以每半个垂体为单位在199培养液中培养(37℃,pH7.3,培养时通以5%CO_2,95%O_2)。在预培养1小时后置换新鲜培养液,经1、2、3小时培养,分别以放射免疫测定法分析培养液中LH含量。用同一垂体前叶之一半与另一半组成一个配对组,观素LH的基础分泌及LHRH促进LH分泌的作用。此外,在培养液中分别加入牛PRL,人工合成精氨酸血管加压素或牛血清白蛋白,培养后测定培养液LH食量,观察PRL,血管加压素及蛋白在离体下对

MATERIALS AND METHODS: Mature female rats were sacrificed by decapitation two weeks after ovariectomy.
