该研究观察抗抑郁中药古方开心散(KXS)在体内外对单胺氧化酶(MAO)活性的影响,从代谢的角度探索KXS升高单胺类神经递质水平的机制。通过在体外酶反应体系和神经胶质细胞C6中分别观察不同浓度的KXS对MAO-A和MAO-B活性的影响;在动物水平的研究中,分别观察不同浓度的KXS灌胃给药1周、2周和3周后,对正常大鼠和孤养结合慢性不可预见性中等强度应激(CMS)模型大鼠脑线粒体MAO-A和MAO-B的活性的影响。结果显示,10g·L-1的KXS在酶反应体系中能显著降低MAO-A和MAO-B的活性;在C6细胞中,0.625~10 g·L-1的KXS对MAOA的活性没有显著影响,但对MAO-B的活性有较明显的抑制作用,且呈浓度依赖关系;KXS作用1,2,3周对正常大鼠脑内MAO-A和MAO-B均没有显著地影响;338 mg·kg-1剂量给药2周和3周后,CMS大鼠脑内MAO-A活性较模型组均有所降低(P<0.05),而KXS作用1,2,3周对MAO-B没有显著地影响。结果表明开心散在体外对MAO-A和MAO-B的活性均有一定的影响,在体内对正常大鼠脑内MAO-A和MAO-B的活性没有影响,对CMS大鼠脑内MAO-A有一定抑制作用。
This study was aimed to investigate the effect of KXS on monoamine oxidase (MAO) activity in vitro and in vivo and to explore the mechanism of KXS increasing monoamine neurotransmitters from the perspective of metabolism. The effects of different concentrations of KXS on the activities of MAO-A and MAO-B were observed in the in vitro enzymatic reaction system and the glial cell C6 respectively. In animal studies, different concentrations of KXS were orally administered for one week , 2 weeks and 3 weeks, respectively, on the activity of MAO-A and MAO-B in the brain of normal rats and orphaned and chronic unpredictable moderate-intensity stress (CMS) rats. The results showed that 10g · L-1 KXS could significantly decrease the activity of MAO-A and MAO-B in the enzyme reaction system. In C6 cells, KXS of 0.625 ~ 10g · L-1 had no significant effect on MAOA activity , But had a significant inhibitory effect on the activity of MAO-B in a concentration-dependent manner. KXS had no significant effect on MAO-A and MAO-B in the normal rat brain at 1, 2 and 3 weeks; 338 mg · The MAO-A activity in the brain of CMS rats was lower than that of the model group (P <0.05) after 2 and 3 weeks of administration of kg-1, but no significant effect on the MAO-B Significantly affected.
