目的优化β-榄香烯纳米聚合物胶束的制备工艺。方法以二硬脂酰磷脂酰乙醇胺聚-乙二醇(DSPE-PEG2000)为纳米聚合物胶束的载体材料,采用薄膜水化法,以包封率为考察指标,通过正交试验考察β-榄香烯与DSPE-PEG2000的质量比、水化时间、水化体积、PBS缓冲液的p H值对制备工艺的影响。结果在β-ELE与DSPE-PEG2000的质量比为1∶10,水化体积为5 m L,水化时间为30 min,所用PBS缓冲液的p H值为7.4时,所制得的纳米聚合物胶束形态规则,大小较为均匀,平均粒径为56 nm,平均包封率为88.83%。结论该制备工艺稳定可行,重复性好,可用于β-榄香烯纳米聚合物胶束的制备。
Objective To optimize the preparation of β-elemene nano-polymer micelles. METHODS: The membrane hydration method was used as the carrier material of the polymer micelles of distearoylphosphatidylethanolamine poly-ethylene glycol (DSPE-PEG2000). The entrapment efficiency was taken as the index of investigation. The β- The mass ratio of elemene to DSPE-PEG2000, the hydration time, the hydration volume and the p H value of PBS buffer on the preparation process. Results The results showed that when the mass ratio of β-ELE to DSPE-PEG2000 was 1:10, the volume of hydration was 5 mL, the hydration time was 30 min and the pH value of PBS buffer was 7.4, The morphology of micelles was uniform and the average particle size was 56 nm. The average encapsulation efficiency was 88.83%. Conclusion The preparation process is stable and feasible with good repeatability and can be used for preparation of β-elemene micelles.
