目的比较棉皮素(gossypetin)和棉皮苷(gossypin)抗氧化、保护DNA及阻断亚硝胺合成的作用。方法通过检测棉皮素和棉皮苷还原力的大小初步比较二者抗氧化性的强弱。采用分光光度法比较二者对超氧阴离子自由基(O-2·)、羟基自由基(·OH)和二苯代苦味酰基自由基(DPPH·)的清除作用。采用化学发光法评价了两种化合物对DNA损伤的保护作用。在体外模拟胃酸环境下,检测了二者阻断亚硝胺合成的能力。结果棉皮素的还原力及清除·OH、DPPH·自由基的能力大于棉皮苷。而棉皮苷具有更加显著的保护DNA免受氧化损伤的作用。棉皮素可以更有效的阻断亚硝胺的合成。结论棉皮素的抗氧化性及阻断亚硝胺合成的能力大于棉皮苷,而棉皮苷可以更有效的抑制机体DNA的氧化损伤。
Objective To compare the effects of gossypetin and gossypin on antioxidation, DNA protection and nitrosamine synthesis inhibition. Methods The strength of antioxidant activity of the two preparations was compared by detecting the reducing power of cotton tannin and cannortin. The scavenging effects on superoxide anion radical (O-2 ·), hydroxyl radical (· OH) and dibenzyl picryl radical (DPPH ·) were compared by spectrophotometry. The chemiluminescence method was used to evaluate the protective effect of two compounds on DNA damage. In vitro simulated gastric acid environment, the two tested the ability of nitrosamine synthesis. Results The reducing power of cottonsetin and the ability to scavenge · OH, DPPH · free radicals were greater than that of naringin. While naringin has a more significant role in protecting DNA from oxidative damage. Naringin can more effectively block the synthesis of nitrosamines. Conclusion The anti-oxidative activity of naringin and the ability to block the synthesis of nitrosamines are greater than that of naringin, while naringin can more effectively inhibit the oxidative DNA damage of the body.
