目的探讨结直肠癌患者血浆中小分子核糖核酸-126(Micro RNA-126,mi R-126)的表达水平,及其与临床病理特征之间的相关性,为结直肠癌患者的早期诊断提供可能。方法收集2014年7月至2016年7月在杭州市红十字会医院就诊的45例结直肠癌住院患者和40例健康体检者的外周血,进行实时定量反转录-聚合酶链反应(q Real-Time PCR)检测血浆mi R-126表达水平,并分析血浆mi R-126表达水平与结直肠患者临床病理特征的相关性。用SPSS 16.0和Graph Pad软件进行统计学分析,非正态分布的计量资料的比较用Mann Whitney U检验,计数资料的比较用χ~2检验,通过受试者工作特征(ROC)曲线分析血浆mi R-126对结直肠患者早期诊断的特异性与敏感度。结果结直肠癌患者血浆mi R-126的中位数为0.77,四分位数为0.12和1.31,显著低于健康体检者3.80(0.19,4.11),差异有统计学意义(P<0.05);结直肠癌患者血浆mi R-126低表达与肿瘤转移、肿瘤分化及分期相关(P<0.01)。ROC曲线分析血浆mi R-126曲线下面积为0.89(95%CI:0.80~0.89),区分结直肠癌患者与健康体检者,其特异性为85%,敏感度为92%。结论结直肠癌患者血浆mi R-126表达水平下调与肿瘤发生及远端转移密切相关,是一个潜在的早期诊断标志物。
Objective To investigate the expression of microRNA-126 (mi R-126) in the plasma of patients with colorectal cancer and its correlation with clinicopathological features, so as to provide a possibility for the early diagnosis of patients with colorectal cancer . Methods Peripheral blood samples of 45 colorectal cancer inpatients and 40 healthy volunteers from July 2014 to July 2016 in Hangzhou Red Cross Hospital were collected for real-time quantitative reverse transcription-polymerase chain reaction (q Real-Time PCR) was used to detect the expression of mi R-126 in plasma and the correlation between the expression of mi R-126 and the clinicopathological features of colorectal cancer. Statistical analysis was performed by using SPSS 16.0 and Graph Pad software. Mann Whitney U test was used to compare the non-normal distribution of measurement data. Chi-square test was used to compare the count data. The receiver operating characteristic (ROC) curve was used to analyze plasma mi Specificity and sensitivity of R-126 in early diagnosis of colorectal cancer. Results The median of mi R-126 in patients with colorectal cancer was 0.77, with a quartile of 0.12 and 1.31, which was significantly lower than 3.80 (0.19 and 4.11) in healthy subjects (P <0.05). The low expression of mi R-126 in patients with colorectal cancer correlated with tumor metastasis, tumor differentiation and staging (P <0.01). The area under the curve of ROC curve for plasma mi R-126 was 0.89 (95% CI: 0.80-0.89). The specificity was 85% for colorectal cancer patients with 92% sensitivity. Conclusion The downregulation of plasma mi R-126 expression in patients with colorectal cancer is closely related to tumorigenesis and distant metastasis, which is a potential early diagnosis marker.
