[目的]探讨恶性肿瘤患者细胞因子诱导的杀伤细胞(CIK)体外培养过程中细胞增殖、淋巴细胞亚群表达与乙肝病毒(HBV)携带状况的关系。[方法]80例肿瘤患者根据HBV表面抗原表达状况分为HBV阴性组和HBV阳性组。分别抽取患者外周血54ml进行CIK细胞诱导培养,在第1、5、7、10、13、15d对细胞进行检测,包括细胞计数、细胞活率和细胞表型分析。[结果]两组患者CIK细胞生长情况细胞数量比较差异无统计学意义,增殖倍数HBV阳性组高于HBV阴性组,并且在第13d和15d差异有统计学意义(P<0.05);细胞亚群分析中CD3+T细胞、CD8+T细胞、CD3+CD8+T细胞、CD3+CD56+T细胞比例均随培养时间延长而升高,CD4+T细胞、CD3+CD4+T细胞随培养时间延长而降低,HBV阳性组更显著;其中CD8+T细胞和CD3+CD4+T细胞在两组间的差异具有统计学意义(P<0.05)。CIK细胞回输后HBV阳性组DNA含量较治疗前显著降低。[结论]HBV表面抗原阳性患者CIK细胞较阴性患者扩增能力更强,回输后可以使HBV DNA含量显著降低。
[Objective] To investigate the relationship between cell proliferation and expression of lymphocyte subsets and carrier of hepatitis B virus (HBV) during cytokine-induced killer cells (CIK) in malignant tumor cells in vitro. [Method] 80 patients with cancer were divided into HBV-negative group and HBV-positive group according to the expression of HBV surface antigen. 54ml of peripheral blood of patients were drawn for CIK cell induction culture, and the cells were detected on the 1st, 5th, 7th, 10th, 13th and 15th day, including cell count, cell viability and cell phenotype analysis. [Results] There were no significant differences in the number of CIK cells between the two groups. The positive cells in multiplication group were higher than those in HBV negative group, and the difference was statistically significant on the 13th and 15th days (P <0.05). The cell subsets In the analysis, the proportion of CD3 + T cells, CD8 + T cells, CD3 + CD8 + T cells and CD3 + CD56 + T cells increased with the prolongation of culture time. CD4 + T cells and CD3 + CD4 + T cells were prolonged with culture time (P <0.05). The positive rate of CD8 + T cells and CD3 + CD4 + T cells in the two groups were statistically significant (P <0.05). The DNA content of HBV positive group after CIK cells were transfused was significantly lower than that before treatment. [Conclusion] CIK cells with positive HBsAg-positive patients have stronger ability to amplify CIK cells than those with negative ones. After transfusion, the content of HBV DNA can be significantly reduced.
