目的 探讨拟除虫菊酯对神经系统毒作用影响及其机制。方法 通过核酸原位杂交技术研究氯氰菊酯 (cyper-methrin,Cy)对中枢神经系统一氧化氮合酶 (nitric oxide synthase,NOS) m RNA水平的影响。结果 Cy(2 0 m g/ kg)腹腔注射染毒 3 h后 ,大鼠海马旁回 CA1~ CA4区锥体细胞层、齿状回颗粒细胞层及大脑皮层 1~ 4分子层神经细胞中神经型 NOSm RNA (n NOS m RNA )水平显著升高 ,对照组仅在海马 CA1、CA 3区及大脑皮层 1~ 2分子层有较低水平的表达 ;而诱导型NOS(i NOS)在正常及 Cy染毒条件下均未检测出。结论 n NOS基因表达增强在拟除虫菊酯神经毒作用机制中可能具有重要作用
Objective To investigate the effect of pyrethroid on the nervous system and its mechanism. Methods The effect of cyper-methrin (Cy) on nitric oxide synthase (mNOS) m RNA level in the central nervous system was studied by in situ hybridization. RESULTS: Neurons in neurons of CA1 ~ CA4 pyramidal cell layer, dentate gyrus granule cell layer and cerebral cortex layer 1 ~ 4 neurons in the parahippocampal CA1 area were exposed to Cy (20 mg / kg) intraperitoneally for 3 h. The level of NOSmRNA in the control group was significantly lower than that in the CA1 and CA3 areas of the hippocampus and the level of 1-2 layers in the cerebral cortex. However, the expression of iNOS in normal and Cy Not detected under exposure conditions. Conclusion The enhancement of nOS gene expression may play an important role in the mechanism of pyrethroid neurotoxicity
