为了简化水稻单倍体应用的培养基,我们在应用马铃薯简化培养基的基础上,结合洪湖湖区莲藕多的特点,于1976年9—10月以莲藕作为代用品,取代当前常用的诱导培养基(米勒培养基)中的全部和部分大量元素、微量元素和有机物质。试验结果表明,用一定浓度的莲藕提取液作为培养基的基本成份,对诱导水稻花粉愈伤组织,提高诱导频率,具有一定的效果。一、简化培养基的制备。利用洪湖湖泊里生长的野藕作莲藕水提液的方法是:称取新鲜野藕顶芽节和芽节近处藕200克,洗净去皮,切成1厘米见方的碎块,加适量蒸馏水煮沸20分钟,用双层纱布过滤,取滤液备用。在滤液中加入铁盐、生长刺激素、蔗糖、琼脂,最后加蒸馏水至1,000毫升,即成含20%莲藕
