目的:建立一种检测枸杞子和大枣中甜菜碱含量的直接离子色谱法。方法:采用Thermofisher Ion Pac CS19(250mm×4mm)阳离子交换色谱柱和CG19(50mm×4mm)阳离子保护柱,以甲基磺酸(MSA)为流动相,等度洗脱;流速1.0m L/min。结果:用该离子色谱法测定枸杞子和大枣中的甜菜碱,在10min内可完成分离检测;甜菜碱质量浓度在10-500mg/L内标准曲线具有良好的线性关系(r2=0.9997);该方法的最低检出限为1.8mg/L;精密度良好,保留时间和峰面积的RSD(n=6)分别为0.15%和1.7%;枸杞子和大枣的加样回收率在85.1%-93.4%之间(n=6),平均加样回收率分别为88.8%和89.7%;样品在室温下放置12h稳定。结论:运用该法测定甜菜碱含量灵敏度高、重现性好、检测过程简便,具有较高的实用和推广价值。
Objective: To establish a direct ion chromatography detection of medlar and jujube betaine content. METHODS: Thermofisher Ion Pac CS19 (250mm × 4mm) cation exchange column and CG19 (50mm × 4mm) cation guard column were used to elute isocratic eluate with methylsulfonic acid (MSA) as mobile phase. The flow rate was 1.0m L / min . Results: The content of betaine in Fructus Lycii and Jujube was determined by ion chromatography in 10 minutes. The calibration curve of betaine with 10-500 mg / L showed good linearity (r2 = 0.9997). The detection limit of this method was 1.8mg / L; the precision was good, the retention time and peak area of RSD (n = 6) were 0.15% and 1.7% respectively; the recovery of wolfberry fruit and jujube was 85.1% -93.4% (n = 6). The average recoveries were 88.8% and 89.7%, respectively. The samples were stable for 12h at room temperature. Conclusion: The determination of betaine content with high sensitivity, good reproducibility, the detection process is simple, with high practical and promotional value.
