利用等电聚焦(IEF)电泳和不连续醋酸尿素聚丙烯酰胺凝胶电泳(NAU-PAGE)对玉米籽粒贮藏蛋白的等电点(pI)和在F1中的遗传表现以及贮藏蛋白在胚和胚乳中的分布进行研究,结果表明:(1)玉米籽粒贮藏蛋白的pI在3.5-8.45范围内,可分离的有39条左右,60%左右的蛋白质属酸性,pI分布在3.50-6.85范围内,40%左右属中性偏碱,pI在6.85-8.45范围内。(2)籽粒贮藏蛋白在F1代呈共显性遗传,没有偏亲现象。所有被测组合在F1代均有互补带,这是区别自交系与杂交种有效的生化依据。(3)蛋白质在胚中百分含量较高,即浓度较高,胚乳中较低。
The isoelectric point (pI) of maize grain storage protein and its genetic expression in F1 were determined by isoelectric focusing (IEF) electrophoresis and discontinuous urea polyacrylamide gel electrophoresis (NAU-PAGE), and the storage proteins in embryos and endosperm The results showed that: (1) The pI of maize grain storage protein was in the range of 3.5-8.45, and about 39 were separable. About 60% of the proteins were acidic and pI was distributed in 3. In the range of 50-6.85, about 40% is neutral and alkaline, and the pI is in the range of 6.85-8.45. All tested combinations have complementary bands in F1 generation, which is an effective biochemical basis for distinguishing inbred lines and hybrids.
