我室以往工作表明间断常压缺氧2及4周大鼠肺β受体减少,其中主要是β_2受体减少。本文在此基础上探讨常压缺氧后大鼠肺肾上腺素能β_2受体mRNA表达的变化。大鼠置常压缺氧舱内(O_2=10%)间断缺氧(6小时/天,6天/周)。右心导管测定平均肺动脉压;在用放射性配基(~3H-DHA)结合法测定β及β_2受体的同时,用聚合酶链反应(PCR)扩增β_2受体mRNA。根据大鼠β_2受体基因序列,设计合成一对引物,在PCR反应时掺入α-~(32)P-dATP,最后进行液体闪烁测量,以特异性条带放射性(cpm)反映β_2受体mRNA水平的高低。结果表
Previous work in our lab showed that there was a decrease in lung β receptors in rats at 2 and 4 weeks of intermittent normobaric hypobaric hypoxia, with a predominant decrease in β 2 receptors. Based on this, we investigated the changes of the expression of adrenergic β_2 receptor mRNA in rats after hypobaric hypoxia.
