目的探讨联合应用抗凝血药物阿加曲班和抗自由基药物依达拉奉,对沙土鼠15分钟前脑缺血后慢性期认知功能的改善作用以及组织形态学改变。方法采用沙土鼠双侧颈动脉结扎法制作15分钟前脑缺血模型。缺血后分别利用依达拉奉、阿加曲班以及两药联合应用进行神经保护治疗并与对照组进行比较。利用“Y”字穿梭箱观察缺血4周后的学习再记忆能力,同时对脑组织进行组织学观察。结果两药联用虽然不能改善慢性期长期记忆能力,但明显减轻了神经损伤并有效改善了短期记忆能力。结论联合应用阿加曲班和依达拉奉,有效抑制沙土鼠15分前脑缺血后的神经细胞进行性损伤,从而改善慢性期认知功能障碍。
Objective To investigate the effect of combined application of anticoagulant argatroban and antiradical drug edaravone on the cognitive function of gerbils after 15 min of forebrain ischemia and histological changes. Methods A 15-minute forebrain ischemia model was made by bilateral carotid artery ligation in gerbils. After ischemia, edaravone, argatroban and the combination of two drugs were used for neuroprotective treatment and compared with the control group. The “Y” word shuttle box was used to observe the learning and memory ability of the rats after 4 weeks of ischemia, and the brain tissues were also observed for histology. Results Although the combination of the two drugs did not improve chronic long-term memory, it obviously reduced nerve damage and short-term memory. Conclusion The combination of argatroban and edaravone effectively inhibits the progressive injury of nerve cells in gerbils 15 minutes after forebrain ischemia and improves the cognitive dysfunction in chronic phase.
