目的建立一种测定乙醇消毒液中乙醇含量的HPLC法。方法以甘油为内标,乙醇和甘油经对甲苯磺酰异氰酸酯(TSIC)衍生化后衍生化产物用HPLC法测定。色谱柱为Agilent C18(150 mm×4.6 mm,5μm),流动相为乙腈-磷酸缓冲液(0.01 mol·L-1KH2PO4,磷酸调p H至2.5)(35∶65),检测波长227 nm,流速1 m L·min-1,柱温25℃。结果在该条件下,乙醇在质量浓度2.5~200 mg·L-1内线性良好(r>0.999 5,n=7)。回收率为97%~103%(n=5),RSD<3%(n=5)。衍生化产物在12 h内稳定。测得样品中乙醇的标示量百分含量为90%~110%。结论经验证,该方法快速、简便、结果准确、重复性好,适合于乙醇消毒液中乙醇的含量测定。
Objective To establish a HPLC method for the determination of ethanol content in ethanol disinfectant. Methods Glycerol was used as internal standard. The derivatized products of ethanol and glycerol derivatized with p-toluenesulfonyl isocyanate (TSIC) were determined by HPLC. The column was Agilent C18 (150 mm × 4.6 mm, 5 μm). The mobile phase consisted of acetonitrile-phosphate buffer (0.01 mol·L-1 KH2PO4, pH = 2.5) (35:65) 1 m L · min-1, column temperature 25 ℃. Results Under this condition, ethanol had a good linearity (r> 0.999 5, n = 7) in the range of 2.5 ~ 200 mg · L-1. The recoveries ranged from 97% to 103% (n = 5) with RSD <3% (n = 5). The derivatized product is stable within 12 h. Measured in the sample of ethanol labeled amount of 90% to 110%. Conclusion The method has been proved to be fast, simple, accurate and reproducible. It is suitable for the determination of ethanol content in ethanol disinfectant solution.
