经氯化锂—脲提取RNA的方法提取钩端螺旋体(钧体)赖型017株的总RNA,用生物素标记的钩体保护基因BMD-3A、BMD-10、问号钩体种特异性基因CPL5x及赖型钩体017株总DNA作为探针,分别与所提取的总RNA进行斑点杂交。结果显示:以上探针均出现了不同强弱的杂交信号,证明了BMD-3A、BMD-10、CPL5x三个特异性基因片段在017株钩体中得到了转录表达,提示其可能分别在钩体免疫保护及钩体种特异性鉴定中具有重要作用。
The total RNA was extracted from 017 strain of Leptospira interrogans by lithium-urea extraction of RNA. The biotin-labeled leptospira (BMD-3A), BMD-10 CPL5x and Leptospira 017 strains were used as probes to carry out dot blot hybridization with the extracted total RNA respectively. The results showed that all the above probes showed hybridization signals with different strengths and weaknesses, and proved that three specific gene fragments of BMD-3A, BMD-10 and CPL5x were transcribed in 017 leptospira, Body immune protection and hook body species-specific identification plays an important role.
