目的:研究益艾康胶囊对奈韦拉平在大鼠体内药动学的影响,为益艾康胶囊在临床合理用药提供依据。方法:实验大鼠分别灌胃给予奈韦拉平和奈韦拉平+益艾康胶囊,并通过LC/MS/MS测定2种给药方式后血浆中奈韦拉平浓度,随后使用Phoenix Win Nonlin 6.1软件计算药动学参数。结果:符合方法学考察要求:血药浓度在5~500 ng·m L-1范围内线性关系良好,日内和日间精密度均<10.0%,提取回收率89.8%~107.0%,稳定性较好,受基质影响较小。由2组药动学主要参数得知MRT(0~t)和MRT(0~∞)均显著升高(P<0.05),Cmax显著降低(P<0.05),其他参数无统计学意义。结论:益艾康胶囊确实对奈韦拉平在大鼠体内药动学产生了影响。
Objective: To study the effect of Yi Yi Kang capsule on the pharmacokinetics of nevirapine in rats, and provide the basis for Yi Yi Kang Capsule in the clinical rational drug use. Methods: Nevirapine and nevirapine + Yikang Capsules were intragastrically administrated in rats, and the concentration of nevirapine in plasma was determined by LC / MS / MS. The pharmacokinetic parameters were calculated by using Phoenix Win Nonlin 6.1 software. Results: In accordance with the methodological study requirements, the linearity was good in the range of 5-500 ng · m L-1, the intra-day and inter-day precision was <10.0%, the recovery was 89.8% -107.0%, the stability was better Well, less affected by the matrix. The main parameters of two groups of pharmacokinetics showed that MRT (0 ~ t) and MRT (0 ~ ∞) were significantly increased (P <0.05), Cmax was significantly decreased (P <0.05), and other parameters were not statistically significant. CONCLUSION: Yikang Capsule did affect the pharmacokinetics of nevirapine in rats.
