目的 通过观察芦荟大黄素对动脉损伤后平滑肌细胞增殖细胞核抗原 (PCNA)基因及蛋白表达产物的作用 ,探讨芦荟大黄素抑制平滑肌细胞增殖的分子作用机制。方法 用 3FFogarty球囊拉栓导管对纯种日本大耳白兔腹主动脉进行球囊内皮剥脱损伤 ,4 8小时后取出腹主动脉中膜进行原代平滑肌细胞培养 ,细胞同步于G0 /G1期后 ,实验组加芦荟大黄素 2 0 μg/ml,对照组加等体积的细胞培养液 ,18小时后 ,分别用RT/PCR、Western杂交法检测药物组与对照组平滑肌细胞PCNA基因mRNA和蛋白表达产物的变化。结果 与对照组比较 ,加用芦荟大黄素以后 ,平滑肌细胞PCNAmRNA和蛋白水平上的表达均显著抑制。结论 芦荟大黄素对平滑肌细胞的增殖抑制作用可能是通过影响PCNA基因的表达实现的。
Objective To investigate the molecular mechanism of aloe-emodin in inhibiting the proliferation of smooth muscle cells by observing the effect of aloe-emodin on the proliferation of PCNA gene and protein expression product after arterial injury. Methods Abdominal aorta of rabbits was isolated from the abdominal aorta of a pure Japanese white rabbit by 3FFogarty balloon embolization catheter. After 48 hours, the abdominal aorta middle membrane was removed to culture the primary smooth muscle cells. The cells were synchronized to the G0/G1 phase. After the experiment group, aloe-emodin 20 μg/ml was added to the control group, and an equal volume of cell culture medium was added to the control group. After 18 hours, the PCNA gene mRNA and protein in the smooth muscle cells of the drug group and the control group were detected by RT/PCR and Western blot respectively. Expression product changes. Results Compared with the control group, the expression of PCNA mRNA and protein in smooth muscle cells was significantly inhibited after adding aloe-emodin. Conclusion The inhibitory effect of aloe-emodin on the proliferation of smooth muscle cells may be achieved through affecting the expression of PCNA gene.
