目的探讨定心方对动脉粥样硬化(AS)小鼠的防治作用及其机制。方法 50只载脂蛋白E基因敲除(ApoE~(-/-))小鼠喂食高脂饲料并随机分为5组:定心方低、中、高剂量组分别给予定心方(DXR)9.375,18.75,37.5 g/(kg·d)灌胃;辛伐他汀组予辛伐他汀5 mg/(kg·d)灌胃;模型组予等剂量生理盐水。另设10只C57BL/6J小鼠喂食普通饲料为对照组。14周后测小鼠血清脂质运载蛋白-2(LCN-2)、肿瘤坏死因子-α(TNF-α)、单核细胞趋化蛋白-1(MCP-1)及白细胞介素-6(IL-6)水平;HE染色观察主动脉组织病理形态学改变;Western blot检测主动脉LCN-2蛋白表达。结果DXR中、高剂量组及辛伐他汀组小鼠血清LCN-2、TNF-α、MCP-1及IL-6水平、主动脉LCN-2蛋白表达及主动脉AS斑块面积均显著低于模型组(P<0.05或P<0.01)。结论 DXR可能通过降低LCN-2表达,抑制炎症反应,显著减轻ApoE~(-/-)小鼠AS病变。
Objective To investigate the preventive and therapeutic effects of Dingxin Recipe on atherosclerosis (AS) mice and its mechanism. Methods 50 ApoE ~ (- / -) mice were fed with high fat diet and randomly divided into 5 groups: DXR group, DXR group, 9.375, 18.75 and 37.5 g / (kg · d) respectively. Simvastatin group was given simvastatin 5 mg / (kg · d) orally. The model group was given normal saline. Another 10 C57BL / 6J mice were fed with normal diet as the control group. Serum lipoxygenin-2, tumor necrosis factor-α (TNF-α), monocyte chemotactic protein-1 (MCP-1) and interleukin-6 IL-6). The pathological changes of the aorta were observed by HE staining. The expression of LCN-2 protein in the aorta was detected by Western blot. Results The serum level of LCN-2, TNF-α, MCP-1 and IL-6, the expression of LCN-2 protein in aorta and the area of aortic AS plaque in DXR high dose and simvastatin groups were significantly lower Model group (P <0.05 or P <0.01). Conclusion DXR may reduce the expression of LCN-2, inhibit the inflammatory reaction and ameliorated the pathological changes of AS in ApoE ~ (- / -) mice.
