目的:考察槐定碱脂质体不同制备方法对其包封率的影响。方法:采用注入、薄膜分散、逆向蒸发等被动载药法和硫酸铵梯度、pH梯度结合逆向等主动载药法制备槐定碱脂质体,用高速冷冻离心-HPLC法测定脂质体包封率。结果:被动载药法制备的脂质体包封率较低,主动载药法中pH梯度结合逆向法制备脂质体包封率为93.27%。结论:采用主动载药法可制得包封率较高的槐定碱脂质体。
OBJECTIVE: To investigate the effects of different formulations of sophoridine liposomes on the entrapment efficiency. METHODS: Sophoridine liposomes were prepared by passive drug loading methods such as injection, membrane dispersion and reverse evaporation, ammonium sulfate gradient, pH gradient and reverse direction, and the liposome encapsulation was determined by high speed freeze-centrifugation-HPLC rate. Results: The entrapment efficiency of liposomes prepared by passive drug loading method was low. The entrapment efficiency of liposomes prepared by pH gradient and reverse method was 93.27%. Conclusion: Sophoridine liposomes with high encapsulation efficiency can be obtained by active drug loading method.
