目的:采用 HPLC 法测定芫荽籽中绿原酸的含量。方法:色谱柱为 Hypersil-ODS 柱(6.0 mm×125 mm,5μm),以甲醇-0.4%乙酸(25:75)为流动相,流速为1.0 mL·min~(-1),柱温为室温,检测波长328 nm,进样量5 μL。结果:在上述色谱条件下,绿原酸对照品的峰面积与其质量在0.015~0.106μg范围内线性关系良好,回归方程为 Y=9903.85X+12.72,r=0.9996,平均回收率为98.3%(n=3)。结论:此方法简单快速,结果准确。
Objective: To determine the chlorogenic acid content in loquat seeds by HPLC. Methods: The column was Hypersil-ODS column (6.0 mm×125 mm, 5 μm). The mobile phase consisted of methanol-0.4% acetic acid (25:75), the flow rate was 1.0 mL·min-1, and the column temperature was room temperature. The detection wavelength is 328 nm and the injection volume is 5 μL. Results: Under the above chromatographic conditions, the linear relationship between the peak area of the chlorogenic acid reference substance and its mass ranged from 0.015 to 0.106 μg, the regression equation was Y=9903.85X+12.72, r=0.9996, and the average recovery was 98.3% ( n=3). Conclusion: This method is simple, fast, and accurate.
