目的建立HPLC测定注射用蒲公英粉针剂中绿原酸、咖啡酸含量的方法。方法采用DikmaTMC18柱(200mm×4.6mm,5μm),流动相为乙腈-0.1%磷酸水溶液(17:83),流速为1.0mL·min-1,检测波长为326nm,柱温为30℃。结果绿原酸浓度0.108~3.464μg·mL-1与峰面积的线性关系良好(r=0.9998),平均加样回收率为99.68%(n=9);咖啡酸浓度0.114~3.648μg·mL-1与峰面积的线性关系良好(r=0.9999),平均加样回收率为98.73%(n=9)。结论所建方法灵敏、简便、准确、重复性好,可用于蒲公英冻干粉针剂中绿原酸、咖啡酸的含量测定。
Objective To establish a HPLC method for the determination of chlorogenic acid and caffeic acid in dandelion injection for injection. Methods The mobile phase was acetonitrile-0.1% phosphoric acid solution (17:83) at a flow rate of 1.0 mL · min-1 using a DikmaTMC18 column (200 mm × 4.6 mm, 5 μm) with a detection wavelength at 326 nm and a column temperature of 30 ° C. Results The linear relationship between the concentration of chlorogenic acid and peak area was 0.108 ~ 3.464μg · mL-1 (r = 0.9998), the average recovery was 99.68% (n = 9). The caffeic acid concentration was 0.114-3.648μg · mL- 1 had a good linear relationship with the peak area (r = 0.9999), and the average recovery was 98.73% (n = 9). Conclusion The method is sensitive, simple, accurate, reproducible and can be used for the determination of chlorogenic acid and caffeic acid in the freeze-dried powder injection of dandelion.
