诱导神经上皮干细胞(induced neuroepithelial stem cells,i NESCs)具有很强的增殖能力和分化潜能,具有重要的科学和临床应用价值。通过i NESCs自体移植产生功能性神经元,在治疗神经性疾病和神经损伤中具有广阔的前景。然而,当前只能通过外源基因过表达的方法获得灵长类i NESCs,这存在未知的安全风险。因此,通过小分子化合物、3D微环境、压力刺激等非外源基因导入法获得灵长类i NESCs显得重要而迫切。长期以来,大量的研究使用啮齿类动物模型来探索药物和细胞在体内的安全性和有效性,然而啮齿类动物与人存在的巨大物种差异导致很多研究在临床试验中失败。因此,借助与人类亲缘关系相近的非人灵长类动物进行研究,进而评估和测试i NESCs在体内的安全性和有效性,是推动人i NESCs进入临床转化更有效的方法。
Induced neuroepithelial stem cells (iNESCs) have strong proliferative capacity and differentiation potential, and have important scientific and clinical value. The generation of functional neurons by autologous transplantation of i NESCs has broad prospects for the treatment of neurological diseases and nerve damage. However, primate i NESCs can only be obtained by overexpression of exogenous genes, which poses an unknown safety risk. Therefore, it is important and urgent to obtain primate i NESCs by introducing non-exogenous gene such as small molecule compounds, 3D microenvironment and pressure stimulation. For a long time, a large number of studies have used rodent models to explore the safety and efficacy of drugs and cells in vivo. However, the huge species differences between rodents and humans lead to many studies failing in clinical trials. Therefore, it is a more effective method to promote the entry of human i NESCs into clinical transformation by studying nonhuman primate with similar genetic relationship to evaluate and test the safety and effectiveness of iNESCs in vivo.
