目的研究归芪聪志汤对血管性痴呆(VD)模型大鼠大脑皮层线粒体功能的影响。方法将120只大鼠称重,进行水迷宫测试,剔除不合格大鼠10只,使用区组随机法选出10只大鼠作为假手术组,其余大鼠采用两血管阻断改良法制备VD大鼠模型,经Morris水迷宫筛选出造模成功的大鼠,随机分为模型组,阳性药物对照组,归芪聪志汤高、中、低剂量组。各组给予相应的药物或蒸馏水灌胃4周后,HE染色观察海马神经细胞的损伤情况,采用比色法检测大鼠海马组织及大脑皮层其他部位线粒体复合物的活性及三磷酸腺苷(ATP)的含量。结果与假手术组比较,模型组海马神经细胞损伤程度严重;与模型组比较,各治疗组海马神经细胞损伤程度均减轻,其中归芪聪志汤中剂量组海马神经细胞损伤程度改善最为明显。与假手术组比较,模型组海马组织及大脑皮层其他部位线粒体复合物Ⅰ~Ⅴ的活性及ATP含量均明显降低(P<0.05)。与模型组比较,阳性药物对照组与归芪聪志汤中剂量组大鼠海马组织线粒体复合物Ⅰ~Ⅴ的活性及ATP含量明显升高,除归芪聪志汤中剂量组的线粒体复合物Ⅰ与模型组比较差异无统计意义外,其他均有统计意义(P<0.05);阳性药物对照组与归芪聪志汤中剂量组大鼠大脑皮层其他部位线粒体复合物Ⅰ~Ⅴ的活性均升高,其中线粒体复合物Ⅰ,Ⅲ,Ⅳ,Ⅴ差异有统计意义(P<0.05),ATP含量均降低,但差异无统计意义(P>0.05)。与阳性药物对照组比较,归芪聪志汤中剂量组大鼠海马组织线粒体复合物Ⅴ的活性明显升高(P<0.05);归芪聪志汤中剂量组大脑皮层其他部位线粒体复合物Ⅰ活性明显升高,线粒体复合物Ⅴ活性明显降低,差异均有统计意义(P<0.05)。结论归芪聪志汤能明显改善VD模型大鼠海马组织病变,其作用机制与改善海马组织和大脑皮层其他部位线粒体复合物Ⅰ~Ⅴ的活性及ATP含量有关。
Objective To study the effects of GuiQiChiZhiTang on mitochondrial function in cerebral cortex of vascular dementia (VD) model rats. Methods 120 rats were weighed, water maze test was performed, 10 unqualified rats were excluded, 10 rats were randomly selected as the sham operation group, and the other rats were prepared by two vascular blockade modified method The rats were screened out by Morris water maze. The rats were randomly divided into model group, positive control group, Gui Zhi Cong Zhi Tang high, medium and low dose groups. The rats in each group were given the corresponding drugs or distilled water for 4 weeks. The damage of hippocampus neurons was observed by HE staining. The activity of mitochondrial complex and the content of adenosine triphosphate (ATP) in hippocampus and other parts of cerebral cortex were detected by colorimetry. . Results Compared with the sham-operation group, the damage of hippocampal neurons in the model group was severe. Compared with the model group, the damage of hippocampal neurons in each treatment group was alleviated. The injury of hippocampus neurons in Guqitongzhi decoction group was most obvious. Compared with the sham operation group, the activity and ATP content of mitochondrial complex Ⅰ-Ⅴ in the hippocampus and other parts of the cerebral cortex in the model group were significantly decreased (P <0.05). Compared with the model group, the activity and ATP content of hippocampus mitochondrial complex Ⅰ-Ⅴ in the positive drug control group and the Guiqu Congzhi decoction medium group were significantly increased, except for the middle dose group (P <0.05). The activities of Ⅰ ~ Ⅴ of mitochondrial complexes in other parts of the cerebral cortex in the positive drug control group and the Guiqi Congzhi decoction group were all significantly lower than those in the model group (P <0.05). The content of ATP in the mitochondrial complex was decreased, but the difference was not statistically significant (P> 0.05). Compared with the positive control group, the activity of mitochondrial complex Ⅴ in hippocampus of Gujing Cong Zhi decoction medium group was significantly increased (P <0.05), while that of Guqi Cong Zhi decoction middle dose group Activity was significantly increased, mitochondrial complex Ⅴ activity was significantly lower, the difference was statistically significant (P <0.05). Conclusion Guqi Cong Zhi Decoction can significantly improve the hippocampal lesion in VD model rats, and its mechanism is related to the improvement of activity and ATP content of mitochondrial complex Ⅰ ~ Ⅴ in hippocampus and other parts of cerebral cortex.
